Ok to run without 1 girdle bolt?

Also with some finesse maybe possible to do without removing cylinder, especially if you're willing to drill the hole in the cylinder out a little over size to clear the major diameter of the tap. Might need long drill bits.

Are you running a type 4 or type 9 pipe? If so I'd hold off on riding unless it's just a stock unported nothing fancy motor.

No, what I should be asking is it ok to run with 1 of the 6 cylinder bolts missing? I am not in a place where I can helicoil or fix it right now

If it's not set up like DAG mentioned and you decide to run it for the weekend, prolly be good to make sure the other 5 are tight.

My Dasa engine had 3 cylinder bolts start to come loose over the whole season, one of them eventually fractured the bolt head off, exhaust side at pto. I noticed it riding one day when adjusting the pipe and noticed one bolt completely backed out, what was left of it.
 
I have rebuilt a 701 top end in the back of my truck at a campground. I would be tearing it down asap to fix rather than waste the funds and time after an issue

If your considering working outta your truck my only recommendation would be drive to somewhere where 27 people don't walk by an hour and ask what you're doing. Really slows the process down.
