You could probably do it with the motor in place, however it would be difficult and you run the risk of making a mistake with having to work in the engine bay (not being able to see under the manifold. You should at least pull the motor to get the manifold installed correctly, the head pipe and...

Rider is a separate issue. We are talking weight of your machine as a whole : Hull and components... A fat dude on the back would do away with any weight savings even on a carbon hull with all the extra time taken to cut weight with components.
